AccuraGen Holdings, founded in 2013, is developing a liquid biopsy technology platform called Firefly, which uses NGS for analysis. The platform is able to identify low-frequency mutations in cfDNA by creating DNA concatemers (long, continuous DNA polymer that contains multiple copies of the same DNA sequence linked in series).
The technology uses rolling circle amplification in which cfDNA is denatured to produce ssDNA fragments. This DNA is amplified via rolling circle amplification (DNA is circularized by ligation reaction). This circularized DNA is then amplified to form a concatemer with multiple tandem repeats. After this step, target enrichment and sequencing are performed.
